Allostera Pharma Inc. is a biopharmaceutical company that has developed an entirely new class of drugs, called Allosteramers™.

Properties of Allosteramers™

Allosteramers™ have three main properties:

  1. they are orally available;
  2. they have predictable and consistent drug properties including their exceptional metabolic profile and significant potency / selectivity similar to that seen with monoclonal antibodies; and
  3. they are “allosteric”, meaning they do not bind to the binding site of the natural ligand the same as traditional drugs, but are engineered to bind at a different site (i.e. by definition an “allosteric” site) potentially providing better safety, efficacy and an entirely new set of patentable intellectual properties.

Allostera’s technology to develop Allosteramers™, called Module X, allows target to lead generation of Allosteramers™ within months rather than the years necessary with other technologies. Using its platform, Allostera has developed a pipeline of seven different Allosteramers™ against different targets primarily focused on inflammation and autoimmunity.

Allostera’s Lead Product

Allostera’s lead Allosteramer™, APG2305, is an orally active drug inhibiting one of the most important targets in the autoimmune disease field, IL23R. IL23 has recently been accepted as the master regulator of TH17 cells, a newly discovered class of T cells which are widely accepted to be the mediators of autoimmune diseases. By shutting off the IL23 receptor, it is believed that most autoimmune diseases, such as psoriasis, rheumatoid arthritis, Crohn’s disease and multiple sclerosis, can be safely and effectively controlled. APG2305 has demonstrated proof-of-concept in animals and is ready to begin IND-enabling studies.

Allostera also has an oral IL1R inhibitor, APG101.10, which has also demonstrated proof-of-concept in animal inflammation models including gout.
